In August 2026, Moderna and Merck announced that their personalized mRNA vaccine — intismeran autogene (previously mRNA-4157 / V940) — combined with the immunotherapy Keytruda, succeeded in a large Phase 3 melanoma trial. What actually happened

At a planned interim analysis, the combination produced a statistically significant improvement in recurrence-free survival (how long patients stayed cancer-free) and in distant metastasis-free survival (staying free of cancer spreading to other organs).

How a "personalized" cancer vaccine works

This is not a one-size-fits-all shot. It's built individually for each patient, and the idea is elegant. When a tumor is removed, doctors sequence it and compare it to the patient's healthy cells to find the mutations that are unique to that person's cancer — flags called neoantigens. Those flags are then encoded into a custom mRNA shot — the same core technology behind the COVID vaccines — which teaches the immune system to recognize and attack up to 34 of those cancer-specific targets.

Shares that sat in the mid-$50s in late July spiked to an intraday high near $177 on the news, one of the biggest single-day moves in the company's history, before falling roughly 25% the next day as early enthusiasm cooled. At least one major bank sharply raised its price target on the back of the data.
